If you are coming from Niagara Falls, you can take the Niagara Parkway to Niagara-on-the-Lake and then follow the signs to Inniskillin.

The winery is located near the Niagara Region Transit bus stop, which connects Niagara-on-the-Lake to other cities in the region.
